Synthesis and Spectroscopic Characterization of Fluorescent Silver Nanoclusters Synthesized on Oligonucleotides Containing a G˗Quadruplex Structure
Abstract
Wprowadzenie miejsc inwersji polarności do fragmentu TBA dwufunkcyjnej matrycy oligonukleotydowej skutkowało otrzymaniem nanoklasterów srebra emitujących w zakresie czerwonym z czasem życia krótszym niż w przypadku niemodyfikowanych matryc TBA.
The utilization of specific DNA sequences as scaffolds for silver nanoclusters (AgNCs) enables the modulation of the optical properties of these systems, and comes from the differential affinity of silver ions toward nucleobases, among which, cytosine bases form the most stable complexes C˗Ag+˗C. An advantage of such syntesized fluorophores relies on their high photostability and easy integration with functional domains, including aptamers and
DNAzymes. As part of the doctoral dissertation, a series of bifunctional DNA probes was developed in which a cytosine-rich domain (C12), serving primarily as a template for silver nanoclusters and responsible for generating a fluorescent signal, was attached to oligonucleotides capable of forming G-quadruplexes (G4), specifically the telomeric sequence (Tel22) and the thrombin-binding aptamer sequence (TBA). Studies employing UV-Vis fluorescence and circular dichroism (CD) spectroscopy demonstrated that the synthesis of silver nanoclusters on these bifunctional DNA constructs does not alter the ability of the G-quadruplex domain to complex K+ ions. A key element of the thesis was the analysis of how oligonucleotide architecture influencesthe probe's analytical performance. In the first stage of the study, it was found that fluorescent silver nanoclusters synthesized on bifunctional oligonucleotides containing G4 Tel22 structure exhibited higher sensitivity to K+ ions than those based on the TBA sequence. It was demonstrated that changing the localization of the C12 domain (from the 3' to the 5' end)
significantly improved emission properties and system stability. Placing the C12 domain upstream of the Tel22 fragment resulted in the formation of silver nanoclusters emitting strong single-band fluorescence in the red region (~620 nm). Other modifications of the C12Tel22 template involved introducing a T5 linker between the C12 and G4 domains or incorporating a fluorescent cytosine analogue, tC. Both approaches yielded highly luminescent systems; however, the T5 linker led to the formation of nanoclusters with green emission (~520 nm), whereas in the second case, tC emission (~510 nm) dominated. Introducing polarity-inversion sites into the TBA fragment of the bifunctional oligonucleotidetemplate resulted in silver nanoclusters that emitted in the red region and exhibited shorter fluorescence lifetimes compared to unmodified TBA templates. Additionally, DLS analysis revealed a tendency for these systems to form aggregates. The red-emitting C12Tel22-AgNCs system demonstrated the highest efficiency in monitoring changes in potassium ion (K+) concentration in the presence of Na+ ions, achieving LOD of 0.68 mM for both the unmodified matrix and the one modified with the cytosine derivative tC. The C12Tel22-AgNCs and C126tCTel22-AgNCs systems exhibit a linear working range for K+ detection from 1 to 10 mM in the presence of 150 mM Na+, fully covering clinically relevant physiological concentrations of potassium ions in extracellular fluids and blood plasma, which typically range from 3.5 to 5.5 mM. Results obtained under conditions
mimicking the extracellular environment, supported by microscopic analyses (AFM, TEM) and cellular studies, confirmed the high application potential of the developed probes for investigating transmembrane potassium ion transport.
